Simplified Antibody quantification using Amperia RED

An automated ‘dip and measure’ approach uses pre-coated Protein A sensors optimised for capture of antibodies. Ideal for antibody quantification or binding assays using an antibody as the capture molecule. The assay takes place directly on the sensor tip, where the bound analyte triggers an electrical signal, which is read instantaneously by the device.

Protein A

A range of pre-coated RED-Sensors for different applications is available. In this application, the Protein A RED-Sensor is selected, enabling Fc-specific antibody capture without the need for sample modification.

The RED-Sensor is dipped into the sample well containing antibody of unknown concentration. Protein A on the sensor surface binds the Fc region of the antibody.

The RED-Sensor is transferred into a detection well containing an HRP-tagged detection antibody. This binds to remaining accessible Protein A sites.

Electrochemical Measurement

The RED-Sensor enters a substrate well containing TMB (Tetramethylbenzidine). Instead of measuring colour change, RED detects the redox reaction electrochemically and in real time. The measured signal is inversely proportional to antibody concentration, and quantification is performed using a standard curve.

After the measurement, sensors regenerate automatically. A new cycle can begin immediately using Steps 2–4. Each Protein A RED-Sensor can be reused for 10 or more quantification iterations.
